The trend towards disposable processing systems in biopharmaceutical manufacturing continues, driven by the considerable economic and operational advantages of these systems. This presentation describes a single-use stirred-tank bioreactor system that has been developed by HyClone Laboratories, Inc., and Baxter Healthcare Corp.
The system consists of a novel single-use container that is held by a matching permanent support vessel and controlled using existing technology. The single-use portion of the system consists of a flexible plastic container with the following single-use integrated components: a bearing, shaft, and impeller assembly; a sparger assembly; ports for sterile attachment of sensor probes; and various ports for inlet and outlet of liquids and gases. This container is held by a permanent support vessel that contains a motor and drive shaft assembly, heat jacket, and openings for inlets, outlets, and probes. The system is operated using standard sensors and controllers that have industry-accepted track records of performance.
The presentation demonstrates the suitability of the single-use bioreactor system for mammalian cell culture. The scaleable mass-transfer characteristics of the single use stirred tank bioreactor are described and cell growth, metabolism and secretion data from batch cultures using 250-liter prototype systems are presented.

The Hayward, CA, facility offers contract manufacturing of mammalian cell derived therapeutic proteins including process development, pilot and clinical scale production. Kurt Kunas has been a senior process development engineer at the Hayward facility for three years. He received his Ph.D. in Chemical Engineering from Rice University in 1990 and B.S. in Chemical Engineering from the University of Michigan in 1984.
